In the northern section, the headline fixture sees last season's Europa League semifinalists Nottingham Forest host fellow top-flight opposition in Leeds United.
Chelsea last won the Carabao Cup in 2015 when a John Terry strike and an own goal by Kyle Walker gave The Blues a 2-0 victory over Mauricio Pochettino's young Spurs team at Wembley. The west London club have lifted the League Cup five times.
Luton booked their spot in the second round with a 3-0 win over Gillingham Town on Saturday.
Spurs' match against Charlton will be the two clubs' first meeting since 2011 when the top-flight club eased to a 3-0 victory in the third round of the FA Cup thanks to a 49th-minute opener from Andros Townsend and a quickfire double from Jermain Defoe.
Tottenham's last Carabao Cup triumph came back in 2008 when Jonathan Woodgate's extra-time goal earned them a 2-1 win over Chelsea.
As was the case for the first round, the second-round draw was regionalised into northern and southern sections, meaning clubs will only be drawn against another team from their respective region.
The regionalisation is used in the opening rounds of the competition to limit travelling distances for clubs and supporters.
Nico O'Reilly's second-half brace against Arsenal at Wembley earned Manchester City their ninth Carabao Cup last season. City's victory meant then-head coach Pep Guardiola became the first manager to win the Carabao Cup five times.
Second-round ties are scheduled to be played during the week commencing Monday, Aug. 24.
